What is the best way to use RGD "Develop Territory"? How do you determine what region to use this card? I've used it to quickly restore regions with low loyality after using Draft, Seize Horses and Requisitions. Are there more effective and better uses for this card?

Same here, I use it to increase loyalty.
You can also use it in region with too low dev level, some decisions do request a min dev level.

Thank you for your reply. Will the use of Develop Territory in an underdeveloped territory along a chain of roads provide any beneift?
For example, I want a rapid route of travel and supply from Paris to Perpignan for use into Spain. All areas have a Major Road, except Mazamet which has a Road, is Civilized at 51 and is Wooded Hills, but it has no City. Can this area have its Road upgraded to a Major Road? Or, what about the LaFerte area? Is there any benefit to developing this area which already contains a Major Road, Cleared at 20 and has Moors, but does not contain a City? Would it benefit to upgrade LaFerte to Civilized?

Not really. The way you are using these decisions is the best way forward.
