Does it neutralize the effect of "South Russian Cossacks", "Siberian Cossacks" and other cossack "regional" traits?

No, it just make sure that the commander inflicts no command penalty on his Cossack units because he is a non-Cossack nationality (a WHI leader is not a COS leader, so he would have a penalty leading COS units....cancelled by this special ability)
