What do postures do for fleets exactly?

not much

Defensive will let your fleet split past other enemies fleets in the same region, if the enemy is in defensive himself, or if you are not detected.
passive will add a +15% chance retreat and work as defensive otherwise. If you are engaged, you will suffer from a battle penalty.
agressive is not yet used, we planned to have the fleet try intercepting others in the adjacent areas with this posture, but it is not yet implemented.

1. So an offensive posture isn't needed to block an enemy port, right?
2. Regarding naval blockades, I suppose transports cannot block enemy ports, right?
3. Are transports considered by the game engine to determine the supply value of regions with ports? If so, should the player move the transports from port to porto or is everything automatic?

1. no
2. supports units are excluded, and transports are supports. So a fleet with only this kind of ships can't blockade.
3. this is automatic, the port give supplies as long it is unblockaded.
There is a small update planned for 1.06 about fleets, to extend their possibilities. I won't give you details, as I don't know how much time it will asks me to finish the feature.
